This question is purely for curiosity, but can the vastus medialis and the vastus lateralis be isolated during exercise? Or not because they both perform the same function of knee extension.

not isolated, but they can certainly be emphasised.

Squats or leg presses with a close stance emphasise the VL, whereas bulgarian split squats and leg extensions where you turn your toes out 45 degrees at the top of the movement will emphasise the VMO.

Chad Waterbury posted this article about about using the step-down exercise to isolate and strengthen the vastus medialis. Although he was using it as a rehab exercise for knee pain, it can be used for weak point training even if no knee pain is present.

I myself sprained my left knee three weeks ago, and I have been using the step-down exercise for about a week. The pain is almost all gone - although in fairness, it was a minor sprain that was healing on its own anyway.
